About This Program
ONO‐4578 is an investigational selective, oral antagonist of prostaglandin E2 receptor 4 (EP4), which is one of the prostaglandin E2 (PGE2) receptors, developed by Ono Pharma.1 PGE2 is widely produced in the body and mediates fever, pain, and inflammation. EP4 is a G‐protein‐coupled receptor for PGE2 that is found in the cell membrane of gastrointestinal epithelial cells, vascular smooth muscle cells, and other cells. Increased expression of PGE2 and EP4 has been observed in cancer and is associated with immunosuppressive activity, including the suppression of T‑cell activation. ONO-4578 is designed to exert antitumor effects by suppressing EP4-mediated effects of PGE2 in advanced or metastatic solid tumors.
ONO-4578 is currently being investigated in combination with nivolumab and chemotherapy in two Phase 2 studies in adults with gastric cancer and adults with colorectal cancer, respectively.
